Noggin Protein, Mouse (CHO)

Noggin protein is an important BMP inhibitor that plays an indispensable role in the neural tube, somite growth, cartilage morphogenesis, and joint formation. Its homodimeric structure promotes significant interactions with GDF5 and possibly GDF6, inhibiting chondrocyte differentiation. Noggin Protein, Mouse (CHO) is the recombinant mouse-derived Noggin protein, expressed in CHO.

Mammalian noggin is remarkably similar in its sequence to Xenopus noggin, and is similarly active in induction assays performed on Xenopus embryo tissues. In the adult mammal, noggin is most notably expressed in the nervous system^[1]. Noggin is a bone morphogenetic protein (BMP) antagonist expressed in Spemann's organizer. Murine Noggin is expressed in condensing cartilage and immature chondrocytes. In mice lacking Noggin, cartilage condensations initiated normally but developed hyperplasia, and initiation of joint development failed^[2]. Noggin protein binds BMP4with high affinity and can abolish BMP4 activity by blocking binding to cognate cell-surface receptors^[3]. Human Noggin (hNoggin) has 205 amino acids (residues 28–232) after removal of its signal sequence (residues 1–27) and is secreted as a glycosylated, covalently linked homodimer. Human Noggin binds with high affinity to hBMP-4 (and with lower affinity to hBMP-7^[4].

Noggin Protein, Mouse (200 ng/mL, 10 days) induces alkaline phosphatase in human mesenchymal stem cell (HMSC), and induces mineralization in HMSC into the osteoblast phenotype^[5].

Noggin Protein, Mouse (17.8 µg/kg/day, intrathecal administration for 2 weeks) promotes the recovery of motor function and the descending corticospinal tract (CST) regeneration in mouse spinal cord injury models^[6].

1.Measured by its ability to inhibit BMP-4-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The ED₅₀ for this effect is ≤0.01 μg/mL in the presence of 30 ng/mL of Recombinant Human BMP-4 (HY-P7007), corresponding to a specific activity is ≥1×10⁵units/mg.
2.Measured by its ability to inhibit BMP-4-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The ED₅₀ for this effect is ≤0.06 μg/mL in the presence of 10 ng/mL of Recombinant Human BMP-4 (HY-P7007), corresponding to a specific activity is ≥1.7×10⁷units/mg.
3.Measured by its ability to inhibit BMP-4-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The ED₅₀ for this effect is 0.02-0.16 μg/mL in the presence of 50 ng/mL of Recombinant Human BMP-4 (HY-P7007).

It is not recommended to reconstitute to a concentration less than 100 μg/mL in ddH₂O. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Stored at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
