MA220607 exhibits potent broad-spectrum antibacterial activity against both sensitive and drug-resistant Gram-positive strains (MIC = 0.062-2 μg/mL) and Gram-negative strains (MIC = 0.5-4 μg/mL)[1].
MA220607 exhibits bactericidal activity against susceptible S. aureus ATCC25923, Methicillin (HY-121544)-resistant S. aureus ATCC43300, B. subtilis ATCC9372, clinically isolated S. aureus, and A. baumannii ATCC19606, with an MBC/MIC ratio ranging from 2 to 4[1].
MA220607 (0-1 μg/mL; 0-24 h) exerts a concentration-dependent bactericidal effect against S. aureus ATCC25923 and inhibits biofilm formation[1].
MA220607 (2.5-10 μg/mL; 30 min) stimulates the polymerization of purified BsFtsZ protein in a concentration-dependent manner[1].
MA220607 (1 μg/mL) disrupts the membrane integrity of S. aureus ATCC25923, thereby causing leakage of intracellular contents[1].

Aerobic bacterial batch culture grows a closed inoculated population in broth or on agar without continuous medium replacement; growth readouts include turbidity/OD for total suspended biomass and colony-forming units for viable cells able to form colonies on agar. OD-based growth curves reflect light scattering by cells, but OD is instrument-, pathlength-, species-, cell-size-, and density-dependent, so OD should be calibrated or interpreted alongside viable counts when quantitative cell density is required.

Transepithelial/transendothelial electrical resistance assay
TEER measures electrical resistance across epithelial or endothelial monolayers cultured on permeable supports, and the readout reflects ionic conductance through the cell barrier, especially the paracellular pathway regulated by junctional integrity. TEER can be measured without destroying the monolayer and is commonly used before or during transport, permeability, barrier-disruption, and barrier-maturation experiments. TEER values are influenced by biological maturation and technical conditions; reported factors include temperature, medium formulation, passage number, electrode geometry, membrane properties, and junctional length during early monolayer maturation. Gram staining of tissue sections is a histochemical technique used to differentiate Gram-positive and Gram-negative bacteria within histological specimens based on differences in bacterial cell wall structure and dye retention, adapted from classical bacteriological Gram staining into tissue-compatible “histological Gram stain” variants. In tissue applications, modifications of the Brown-Hopps and Brown-Brenn methods are commonly used to improve differentiation of microorganisms embedded within host connective tissue and to reduce overstaining or loss of Gram-negative signal, which are known limitations of earlier approaches. The principle relies on crystal violet-iodine complex retention in Gram-positive organisms and subsequent decolorization and counterstaining steps that allow contrast visualization of Gram-negative organisms against tissue background.
