DPPE-PEG2000-pPB

DPPE-PEG2000-pPB is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against PDGFRβ on the surface of activated hepatic stellate cells, pericytes, and tumor-associated fibroblasts.
Composition and role of each component:
1. DPPE (dipalmitoylphosphatidylethanolamine): Lipid anchoring group-inserts into the lipid bilayer of liposomes/LNPs, providing stable membrane anchorage; its saturated C16 chains confer bilayer rigidity.
2. PEG (polyethylene glycol): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. pPB (platelet-derived growth factor receptor β-binding peptide (pPB)): Targeting ligand-a peptide that specifically recognizes and binds to platelet-derived growth factor receptor beta (PDGFRβ), which is highly expressed on activated hepatic stellate cells (HSCs), pericytes, and tumor-associated fibroblasts. Enables targeted delivery to fibrotic tissues and tumor stroma.
