Sibeprenlimab (VIS649) is a humanized IgG2 monoclonal antibody which inhibits a proliferation-inducing ligand (APRIL). Sibeprenlimab suppresses pathogenic immunoglobulins (IgA and IgM), while preserving antibody responses to mRNA-based vaccines against SARS-COV-2. Sibeprenlimab reduces urinary protein-to-creatinine ratio (UPCR) and glomerular filtration rate (GFR). Sibeprenlimab is promising for the research of IgA nephropathy (IgAN).

    Immobilized APRIL/TNFSF13 Trimer Protein, Human (HEK293, His-Flag, HY-P70733A) can bind Sibeprenlimab. The EC50 for this effect is 5.059 ng/mL.

    Flow cytometric analysis of 1X106 A549 cells with Sibeprenlimab (HY-P99901, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/200 dilution for an hour at 4℃. AF 488-conjugated AffiniPure Goat Anti-Human IgG H&L (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG2 kappa (HY-P99002, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
