LCP
LCP is a polar-responsive fluorescent probe for the cellular microenvironment. LCP responds to polarity changes in the cellular microenvironment via fluorescence resonance energy transfer, emitting blue fluorescence in low-polarity environments (Ex/Em = 420 nm/550 nm) and red fluorescence (Ex/Em = 500 nm/662 nm) in high-polarity environments. LCP enables dual-color visualization of dynamic changes in lysosomes and cytoplasmic membranes during cell apoptosis, and monitors cell viability through localization and emission color changes. LCP can be used in cancer research.

Guidelines (The following are our recommended protocols; this is a guideline and should be modified according to your specific needs).
1. Stock Solution Preparation
1.1 Solvent: Anhydrous methanol.
1.2 Concentration Recommendation: Generally, a high concentration of 1-10 mM stock solution is recommended.
1.3 Storage Conditions: Store at -20 ℃ or -80 ℃ protected from light, and avoid repeated freeze-thaw cycles.
2. Working Solution Preparation
2.1 Diluent: Dilute with PBS.
2.2 Working Concentration: 1-10 μM
Note: Please adjust the dye working solution concentration according to your actual situation, and prepare fresh before use.
3. Cell Staining
3.1 Culture cells on sterile coverslips.
3.2 Remove the coverslips from the culture medium and aspirate excess medium.
3.3 Add the working solution, gently shake to completely cover the cells, and incubate at room temperature for 10 minutes.
3.4 Remove the dye working solution, wash 2-3 times with culture medium, 5 minutes each time, and observe using a fluorescence microscope or flow cytometry.
Note: If flow cytometry is required, the cells should be resuspended with trypsin before staining.
LCP (10 μM) distinguishes between lipid phases in cell-free GUV membrane models by emitting red fluorescence in Ld (high-polarity) phases and blue fluorescence in Lo (low-polarity) phases[1].
LCP has low cytotoxicity to HeLa cells at concentrations up to 40 μM, making it suitable for live cell imaging applications[1].
LCP (10 μM; 15 min) localizes to lysosomes in live HeLa cells (with a colocalization coefficient of 0.91 with LysoTracker Red (HY-D1300)) and to the plasma membrane in Cisplatin (HY-17394)-induced apoptotic HeLa cells (with colocalization coefficients of 0.83 and 0.88 with DiO (HY-D0969))[1].
LCP (10 μM; 10 min) shifts from blue (lysosomal) to red (plasma membrane) fluorescence in a concentration-dependent manner in Cisplatin-induced apoptotic HeLa cells, with blue fluorescence weakening and red fluorescence strengthening as Cisplatin concentration increases[1].
