Amorphin
Amorphin is a phosphorylase b with glycogen metabolism activity. Amorphin binds directly to alpha-actinin, with alpha-actinin mediating its binding to actin filaments. Amorphin can be used for the research of McArdle’s disease and breast cancer.

Biological Activity
Amorphin (Compound 7) (68 h) selectively inhibits the proliferation of MCF-7 human breast cancer cells with an IC50 of 0.95 μM, and has no effect on HCT-116 human colorectal cancer cells at concentrations up to 50 μM[1].
Amorphin (1 μg/mL; 2 h at 4°C) directly binds to biotinylated alpha-actinin in a solid-phase dot blot assay[2].
Amorphin (0.31-20 μg/mL; 15 min) binds with higher affinity to purified alpha-actinin than to purified F-actin in a solid-phase radioimmune assay[2].
Amorphin (0.5 mg/mL; 5 min) binds to actin filaments only when they are decorated with purified alpha-actinin, as observed via negative stain electron microscopy[2].
Amorphin binds specifically to the Z-bands of extracted glycerinated adult chicken pectoralis muscle myofibrils, restoring their electron-dense amorphous structure[2].
Amorphin partial peptide sequencing confirms it is nearly identical to rabbit phosphorylase b, demonstrating amorphin is a phosphorylase b[2].
