Neuro-DiO 4-chlorobenzenesulfonate is a hydrophobic C18 alkyl chain carbocyanine dye with green fluorescence, commonly used as a vascular marker, cell internalizer and deposition agent. Neuro-DiO 4-chlorobenzenesulfonate inserts its alkyl chain into the endothelial plasma membrane via liposome-mediated perfusion to achieve vascular labeling. Neuro-DiO chlorobenzenesulfonate can also stain the cell membrane and cytoplasm of cancer cells to assist in confocal microscopy observations. Neuro-DiO chlorobenzenesulfonate can be released from nanosponges and accumulate on the surface of mouse retina, then internalize into retinal ganglion cells, which is applicable to researches related to glaucoma and other diseases. It should be noted that during liposome-mediated vascular staining in mice, Neuro-DiO 4-chlorobenzenesulfonate may cause leakage of airway lavage fluid.

In Vitro

Neuro-DiO 4-chlorobenzenesulfonate (5 μg/mL) effectively stains the membranes and cytoplasm of C6 rat brain glioma cells and MCF-7 human breast adenocarcinoma cells for use as a counterstain in confocal microscopy studies of lipid nanoparticle localization[2].

In Vivo

Neuro-DiO 4-chlorobenzenesulfonate (1 μL of 50 nm nanosponges loaded with Neuro-DiO; intravitreal injection; both eyes) leads to a time-dependent increase in retinal deposition of Neuro-DiO (reaching up to 71.4% retinal surface coverage by 28 days post-injection) and internalization of the dye by retinal ganglion cells[3].

Dosage: 1 μL of 50 nm nanosponges loaded with Neuro-DiO (10.6 weight % load, 0.58 mg/mL concentration)
Administration: Intravitreal injection; both eyes
Result: Reached 19.6% to 28.5% of retinal surface coverage at 3 days post-injection.
Reached 25.6% to 52.1% of retinal surface coverage at 7 to 14 days post-injection.
Reached 60.2% to 71.4% of retinal surface coverage at 28 days post-injection.
Colocalized with phosphorylated neurofilament-positive retinal ganglion cells at 1 week post-injection, with orthogonal projections confirming internalization within retinal ganglion cell somas.
