NHE-1-IN-1
NHE-1-IN-1 is a non-acylguanidine NHE-1 inhibitor with IC50 values of 0.0065 μM and 9 μM for human NHE-1 and NHE-2, respectively. NHE-1-IN-1 inhibits NHE-1 function by suppressing NHE-1-mediated Na+-dependent intracellular pH recovery after acidification. NHE-1-IN-1 can be used for research on NHE-1-related cardiovascular regulation.

In Vitro
NHE-1-IN-1 (compound 9t) inhibits sodium-dependent pH recovery after acidification in AP1 cells expressing human NHE-1, with an IC50 of 0.0065 μM[1].
NHE-1-IN-1 inhibits NHE-2 activity in AP1 cells expressing human NHE-2 with an IC50 of 9 μM and exhibits 1400-fold selectivity over NHE-1[1].

Cell Cytotoxicity Assay
Cytotoxicity assays are usually based on the assessment of cell membrane damage, which can also be indirectly detected by measuring cell viability. Detection methods include MTT assay, CKK-8 assay, LDH assay and ATP assay, etc.
