PROTAC BCR-ABL Degrader-1 is a PROTAC degrader targeting BCR-ABL, and also selectively inhibits cancer cell proliferation. PROTAC BCR-ABL Degrader-1 induces degradation via the ubiquitin-proteasome system to reduce protein levels. PROTAC BCR-ABL Degrader-1 can be used for research on chronic myeloid leukemia.

PROTAC BCR-ABL Degrader-1 (Protac 1) (0.1-100 μM; 48 h) potently inhibits the proliferation of K562 cells with an IC50 of 0.62 μM, while it shows low cytotoxicity against normal human NCM460 cells with an IC50 > 100 μM[1].
PROTAC BCR-ABL Degrader-1 (1-20 μM; 48 h) reduces the protein levels of BCR-ABL and c-ABL in K562 cells in a dose-dependent manner[1].
PROTAC BCR-ABL Degrader-1 (10 μM; 4-48 h) reduces the protein levels of BCR-ABL and c-ABL in K562 cells in a time-dependent manner[1].
PROTAC BCR-ABL Degrader-1 (10 μM; 48 h following 1 h pre-incubation with proteasome inhibitors) induces the degradation of BCR-ABL in K562 cells in a ubiquitin-proteasome-dependent manner, which is supported by the finding that the proteasome inhibitors MG132 (HY-13259) and Bortezomib (HY-10227) attenuate this degradation effect[1].

- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

This protocol yields a clear solution of 2.5 mg/mL.
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
