PRKD3/PKD3

PKD3, encoded by PRKD3 and also designated PKCν, is the most widely expressed member of the protein kinase D family, with high levels in hematopoietic and immune cells. Activation follows diacylglycerol binding to its C1 domains and phosphorylation by PKCη and other novel PKCs. PKD3 controls cell migration through RhoA and ROCK signaling, regulates mitosis and cytokinesis, and influences Golgi organization and integrin-mediated adhesion. In lymphocytes and monocytes it shapes immune responses, and altered PKD3 activity has been reported in glioma, leukemia and metastatic breast cancer. Its dual role in proliferation and motility makes it an attractive oncology target.
