Description

(R)-Chloro-butylphthalide-O-NBP, a chiral 3-n-butylphthalide derivative, is a neuroprotective agent. (R)-Chloro-butylphthalide-O-NBP shows potent blood-brain barrier (BBB) protective function. In vivo, (R)-Chloro-butylphthalide-O-NBP significantly reduces infarct volume and markedly preserves BBB integrity. (R)-Chloro-butylphthalide-O-NBP can be used for the research of ischemic stroke[1].

In Vitro

(R)-Chloro-butylphthalide-O-NBP (compound R-30) (0.1 μM, 1 μM, 10 μM) demonstrates significant improvement in PC12 cell survival under OGD/R conditions[1].
(R)-Chloro-butylphthalide-O-NBP (0.1 μM, 1 μM, 10 μM) significantly reduces LDH release in OGD/R-injured PC12 cells[1].

In Vivo

(R)-Chloro-butylphthalide-O-NBP (compound R-30) (5-20 mg/kg, i.v.) reduces infarct volume, improves neurobehavioral scores, and protects BBB integrity in a rat MCAO model of ischemic stroke[1].
In rats treated with (R)-Chloro-butylphthalide-O-NBP (i.v.; 100-200 mg/kg; once), body weight, food consumption, postmortem organ weight, and biochemical indicators remained normal, with no significant abnormalities observed[1].

Animal Model: Sprague-Dawley (SD) rats (middle cerebral artery occlusion (MCAO) model)[1]
