Procedure
1. Dilute 7-amido-4-methylcoumarin in assay buffer to concentrations of 0, 0.2, 0.39, 0.78, 1.56, 3.13, 6.25, 12.5, 25, 50 μmol/L. Add 100 μL of each concentration to the wells to initiate the reaction, with three replicates per concentration. Read the fluorescence in kinetic mode for 5 minutes with excitation and emission wavelengths of 380 nm and 460 nm (top read) . Plot the standard concentrations on the x-axis and the measured RFU on the y-axis to create the standard curve and obtain the standard curve equation.
2. Dilute Mouse Aminopeptidase A to 0.4 μg/mL in assay buffer.
3. Dilute the substrate to 200 μM in assay buffer.
4. Add 100 μL of 0.4 μg/mL Mouse Aminopeptidase A and 200 μM substrate to the black 96-well plate. Include a control with assay buffer and substrate only. Perform three replicates for each condition.
5. Read the RFU values in kinetic mode for 5 minutes with excitation and emission wavelengths of 380 nm and 460 nm, respectively.
6. Calculate specific activity:
