ATG5 Protein, Human

Cat. No.: HY-P7620
COA Handling Instructions

ATG5 is an important participant in the formation of autophagy vesicles and binds to ATG12 through a ubiquitin-like system, which is critical for the lipidation of ATG8 family proteins. It plays a crucial role in mitochondrial quality control, affects cell longevity, and is critical for lymphocyte development, antigen processing, axonal morphology maintenance, adipocyte differentiation, primary ciliogenesis, and apoptotic processes. ATG5 Protein, Human is the recombinant human-derived ATG5 protein, expressed by E. coli , with tag free. The total length of ATG5 Protein, Human is 275 a.a., with molecular weight of ~33.0 kDa.

Background

ATG5, a key participant in autophagic vesicle formation, undergoes conjugation with ATG12 through a ubiquitin-like system involving ATG7 as an E1-like activating enzyme and ATG10 as an E2-like conjugating enzyme. The resulting ATG12-ATG5 conjugate serves as an E3-like enzyme essential for lipidation of ATG8 family proteins and their association with vesicle membranes. Beyond its role in autophagy, ATG5 contributes to mitochondrial quality control post-oxidative damage, impacting cellular longevity. It plays a critical role in lymphocyte development, vital for the survival and proliferation of both B and T lymphocytes, and is necessary for optimal antigen processing and presentation for MHC II. Additionally, ATG5 is involved in maintaining axon morphology, normal adipocyte differentiation, and promoting primary ciliogenesis through autophagic pathways. Moreover, it may play a crucial role in apoptotic processes, occurring downstream of caspase activity, and participates in IFN-gamma-induced autophagic cell death through interaction with FADD.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

  • Dilution Calculator

  • Specific Activity Calculator

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration
= ÷

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
