Background

Claudin-4 (CLDN4) protein, a channel-forming tight junction protein, serves as a pivotal mediator of paracellular chloride transport in the kidney, particularly influencing the critical process of paracellular chloride reabsorption in the collecting ducts. Within tight junctions, claudins, including CLDN4, play a crucial role in the specific obliteration of intercellular spaces, exhibiting calcium-independent cell-adhesion activity. This protein establishes direct interactions with key tight junction components such as TJP1/ZO-1, TJP2/ZO-2, and TJP3/ZO-3. Additionally, CLDN4 engages with EPHA2, where EPHA2-mediated phosphorylation of CLDN4 may contribute to the regulation of tight junctions. Notably, CLDN4 also interacts with other claudin family members, such as CLDN1 and CLDN8, further emphasizing its involvement in complex tight junction networks.
