Furin/PCSK3 protein is an endoprotease with RX(K/R)R cleavage activity and is widely expressed in the constitutive secretory pathway.It plays a crucial role in processing various substrates, including TGFB1, and activating TGF-β-1.Furin/PCSK3 Protein, Mouse (HEK293, His) is the recombinant mouse-derived Furin/PCSK3 protein, expressed by HEK293 , with C-10*His labeled tag.

Background

Furin/PCSK3 protein is a widely expressed endoprotease present in constitutive secretory pathways that exhibits cleavage activity at the RX(K/R)R consensus motif. It plays a crucial role in the processing of various substrates, including TGFB1, contributing to the activation of TGF-beta-1. Additionally, Furin/PCSK3 protein is responsible for converting the non-functional prohormone Brain natriuretic factor into its active hormone BNP(1-45) through proteolytic cleavage. Moreover, it regulates the acidification of dense-core secretory granules in islets of Langerhans cells by mediating the processing of the accessory subunit ATP6AP1/Ac45 of the V-ATPase.

Verified Bioactivity

1.Measured by its ability to cleave the fluorogenic peptide substrate pERTKR-AMC. The specific activity is 307.98 pmol/min/µg, as measured under the described conditions.
2.Measured by its ability to cleave 50µM fluorogenic peptide substrate Pyr-Arg-Thr-Lys-Arg-AMC that incubate at room temperature for 5minutes.The specific activity is 1653.016 pmol/min/µg.
