Background

Histone chaperone ASF1A/ASF1A Protein, a pivotal player in chromatin dynamics, facilitates histone deposition, and orchestrates histone exchange and removal during nucleosome assembly and disassembly. Working in collaboration with chromatin assembly factor 1 (CAF-1), ASF1A promotes replication-dependent chromatin assembly, and in conjunction with HIRA, it contributes to replication-independent chromatin assembly. Notably, ASF1A plays a role in the homologous recombination-mediated repair of double-strand breaks, acting by mediating histone replacement at the breaks and facilitating the recruitment of the MMS22L-TONSL complex and subsequent RAD51 loading. Furthermore, ASF1A is involved in the nuclear import of the histone H3-H4 dimer alongside importin-4 (IPO4), specifically recognizing and binding newly synthesized histones marked with H3K9me1K14ac and H4K5K12ac in the cytosol. Essential for the formation of senescence-associated heterochromatin foci (SAHF) and efficient senescence-associated cell cycle exit, ASF1A interacts with histones H3 and H4, as well as with various partner proteins such as CHAF1A, CHAF1B, RBBP4, CABIN1, HAT1, HIRA, NASP, TAF1, UBN1, CDAN1, IPO4, and CREBBP.
