KDM5A is a histone demethylase that specifically targets histone H3 at 'Lys-4', playing a pivotal role in histone code regulation. It does not act on H3 'Lys-9', 'Lys-27', 'Lys-36', 'Lys-79' or H4 'Lys-20', and only demethylates trimethylated/dimethylated (not monomethylated) H3 'Lys-4'. It regulates gene transcription by binding to 5'-CCGCCC-3' DNA motifs, potentially enhancing nuclear receptor-mediated transcription and modulating Hox protein expression during cell differentiation. KDM5A also represses cytokine transcription (e.g., CXCL12) and maintains circadian rhythm periodicity. In a demethylase-independent manner, it coactivates CLOCK-BMAL1-driven transcription of PER1/2 and clock-controlled genes by inhibiting HDAC1 to increase histone acetylation at PER1/2 promoters (By similarity). Evidence suggests it may act as a corepressor for genes like MT1F and promote cancer cell proliferation.
