Background

RANKL/TNFSF11 is a cytokine that binds to TNFRSF11B/OPG and TNFRSF11A/RANK, functioning as an osteoclast differentiation and activation factor. It enhances the ability of dendritic cells to stimulate naive T-cell proliferation and may serve as a key regulator of T-cell and dendritic cell interactions, potentially influencing T-cell-dependent immune responses. Additionally, RANKL/TNFSF11 induces osteoclastogenesis by activating multiple signaling pathways in osteoclast precursor cells, notably triggering sustained oscillations in intracellular Ca2+ concentration, leading to NFATC1 activation and osteoclast-specific gene transcription. During osteoclast differentiation, it activates CREB1 and generates mitochondrial ROS in a TMEM64- and ATP2A2-dependent manner, essential for proper osteoclast formation. By similarity, it may also play a role in increased bone resorption in humoral hypercalcemia of malignancy.
