Background

Ribonuclease UK114/HRSP12 protein plays a crucial role in cellular metabolism by catalyzing the hydrolytic deamination of enamine/imine intermediates that arise during normal metabolic processes. This enzymatic activity is thought to facilitate the safe release of ammonia from these potentially toxic reactive metabolites, thereby mitigating their impact on cellular components. HRSP12 is particularly associated with enamine/imine intermediates formed by various pyridoxal-5'-phosphate-dependent dehydratases, including L-threonine dehydratase. Moreover, it contributes to endoribonucleolytic cleavage of specific transcripts by promoting the recruitment of the ribonuclease P/MRP complex. Acting as a bridge between YTHDF2 and the ribonuclease P/MRP complex, HRSP12 binds to N6-methyladenosine (m6A)-containing mRNAs featuring a 5'-GGUUC-3' motif. The cooperative binding of HRSP12 and YTHDF2 to such transcripts leads to the recruitment of the ribonuclease P/MRP complex, facilitating subsequent endoribonucleolytic cleavage.
