Description

RIPK1 Protein, Mouse (C-His) is the recombinant mouse-derived RIPK1 protein, expressed by E. coli, with C-6*His tag.

Background

p-RIPK1 is a serine-threonine kinase that serves as a key regulator of TNF-mediated apoptosis, necroptosis, and inflammatory pathways. It exhibits dual kinase-dependent and -independent functions: its kinase activity governs cell death (e.g., by forming complex IIa for apoptosis and complex IIb for necroptosis), while its scaffold role promotes cell survival via recruitment to the TNF-R1 signaling complex (TNF-RSC/complex I) and activation of the canonical NF-kappa-B pathway. Under physiological conditions, p-RIPK1 inhibits RIPK3-dependent necroptosis (through FADD-mediated CASP8 recruitment to cleave RIPK1) and aberrant apoptosis during embryogenesis (by blocking TRADD-FADD interaction). It also drives inflammatory responses by enhancing pro-inflammatory cytokine (e.g., IL6) production. By similarity, it reciprocally phosphorylates RIPK3 and phosphorylates DAB2IP at Ser-728 in a TNF-alpha-dependent manner to activate the MAP3K5-JNK apoptotic cascade.

Reconstitution
