Background

Sialic acid-binding immunoglobulin-like lectin 5 (Siglec-5) is a member of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. Siglec-5 is a member of the CD33-related subset of Siglecs and is expressed on myeloid cells of the hemopoietic system. Siglec-5 inhibits the response of innate immune cells, such as monocytes and neutrophils against pathogens. Overexpression of Siglec-5 inhibits the activity of T cell receptor (TCR)-induced transcription factors. In addition, binding of Siglec-5 to the GBS β-protein reduces primary T cell activation and leads to decreased production of cytokines and surface antigens. A soluble form of Siglec-5 enhances cytokine production and cytotoxic granule release by cancer antigen-specific T cells. Siglec-5 is an inhibitory receptor with the potential to mediate SHP-1 and/or SHP-2-dependent signaling in the absence of tyrosine phosphorylation. Plasma Siglec-5 can be used as a diagnostic marker for critical Limb Ischemia and fulminant myocarditis[1][2][3][4].
