SIRP alpha V8/CD172a Protein, Human (AAH38510, HEK293, His) is the recombinant human-derived SIRP alpha V8/CD172a protein, expressed by HEK293, with C-His labeled tag.

Background

SIRP alpha V8/CD172a is an immunoglobulin-like cell surface receptor that primarily binds to CD47. It acts as a docking protein, facilitating the translocation of PTPN6, PTPN11, and other binding partners from the cytosol to the plasma membrane, and supports cerebellar neuron adhesion, neurite outgrowth, and glial cell attachment. This protein may play a key role in intracellular signaling during synaptogenesis and synaptic function (By similarity). Additionally, it negatively regulates receptor tyrosine kinase-coupled cellular responses induced by cell adhesion, growth factors, or insulin, inhibiting phagocytosis, mast cell activation, and dendritic cell activation. Binding to CD47 prevents the maturation of immature dendritic cells and suppresses cytokine production by mature dendritic cells. The protein contributes to antiviral immunity by limiting New World arenavirus infection through reduced virus internalization (By similarity). It also serves as a receptor for THBS1, with their interaction stimulating SIRPA phosphorylation (By similarity). In response to THBS1, it participates in ROS signaling in non-phagocytic cells, promoting NADPH oxidase-derived ROS production.

Biological Activity

Immobilized human CD47 at 2 μg/ml (100 μl/well) can bind human SIRPa(V8)-His, the EC50 of human SIRPa(V8)-His is 237 ng/mL.
