Arabinoxylan Medium viscosity is an orally active Dectin-1 splice variant modulator, glucose absorption inhibitor, and chyme viscosity enhancer. Arabinoxylan Medium viscosity inhibits particulate β-glucan-induced Dectin-1A activation and mildly suppresses Dectin-1B activation. In human dendritic cells stimulated with particulate β-glucan, Arabinoxylan Medium viscosity reduces the production of IL-10 and TNF-α, and increases the production of IL-4 and IL-23. Arabinoxylan Medium viscosity also supports antifungal immune responses without activating TLR2, TLR4 or TLR5, and does not induce cytokine production when used to stimulate human dendritic cells alone. Arabinoxylan Medium viscosity increases small intestinal chyme viscosity, gets degraded in the large intestine to produce short-chain fatty acids, reduces glucose absorption and insulin response, and improves glucose homeostasis. Arabinoxylan Medium viscosity supports microbial fermentation and the growth of beneficial microbiota in the gastrointestinal tract, prevents bile acid reabsorption, and delays starch digestion. Arabinoxylan Medium viscosity can be used in research related to type 2 diabetes, impaired glucose tolerance, and metabolic syndrome .

Toll-like Receptor (TLR) MyD88 NF-κB Interleukin Related IFNAR Inflammation/Immunology Cancer
Flagellin from S. typhimurium is a potent TLR5 agonist. Flagellin from S. typhimurium activates immune cells and inhibits the activity of melanoma cells. Flagellin from S. typhimurium activates the NF-κB pathway dependent on the TLR5/MyD88/TRAF6 signaling axis in cells. Flagellin from S. typhimurium induces a proinflammatory response in the primary chicken hepatocyte-nonparenchymal cell co-culture system by promoting IL-8 production, inhibiting IL-10 production, and increasing the IFN-γ/IL-10 ratio. Flagellin from S. typhimurium can be used for research on melanoma and inflammatory diseases .

    mRNA Inflammation/Immunology
Human TLR5 mRNA encodes the human toll like receptor 5 (TLR5) protein, a member of the Toll-like receptor (TLR) family which plays a fundamental role in pathogen recognition and activation of innate immunity. TLR5 recognizes bacterial flagellin, the principal component of bacterial flagella and a virulence factor. The activation of this receptor mobilizes the nuclear factor NF-kappaB, which in turn activates a host of inflammatory-related target genes.

ETI41 is an orally active, selective TLR inhibitor that targets the nucleoside-binding Site I on TLR7 (IC50 = 0.63 μM) and TLR9 (IC50 = 0.16 μM), sparing surface TLRs (including TLR1/TLR2, TLR2/TLR6, TLR4 and TLR5). ETI41 potently inhibits endosomal TLR-mediated pro-inflammatory signaling with nanomolar activity in cellular, biophysical and in vivo assays. ETI41 suppresses the expression of inflammation-associated genes and effectively ameliorates symptoms in mouse models of psoriasis, and systemic lupus erythematosus (SLE). ETI41 can be used for autoimmune and inflammatory diseases research .

Bacterial Toll-like Receptor (TLR) Collagen Infection Inflammation/Immunology
Antibacterial agent 284 (Compound 7) is an Antibacterial agent. Antibacterial agent 284 has a zinc-binding structure and potent inhibitory activity against Legionella pneumophila metalloprotease ProA (IC50: 0.96 μM) with zinc-binding structure. Antibacterial agent 284 significantly inactivates the cleavage of collagen IV and flagellin (ProA substrates) and reduces immune evasion from the TLR5-NF-κB pathway and PMN-mediated inflammation in human lung tissue explants. Antibacterial agent 284 is promising for Legionnaires' disease research .
