TAB-004 is a murine IgG1 monoclonal antibody that specifically binds to the tumor-associated hypoglycosylated mucin 1 (tMUC1), with high selectivity for human tMUC1. TAB-004 can be conjugated with Indocyanine green (ICG) (HY-D0711) for in vivo targeted imaging. TAB-004 (30 min) specifically binds to human tMUC1-expressing MMT murine breast cancer cells, with no detectable binding to tMUC1-negative PyMT murine breast cancer cells[1].
TAB-004 (3 ng/ml) potently and specifically binds to the MUC1 peptide with detectable binding at 20 pM in a cell-free ELISA[2].
TAB-004 specifically detects human MUC1 in human and murine pancreatic cancer cell lines (KCM, BxPC3.MUC1, HPAF-II) via Western blot, with no cross-reactivity to murine MUC1 in KCKO cells[2].
TAB-004 specifically detects the expression of tMUC1 in BxPC3, PANC-1 and HPAF-II human pancreatic cancer cells by flow cytometry, and can recognize tMUC1 on the surface of more than 95% of Triple+ and CD133+ pancreatic cancer stem cells[2].

In Vivo

TAB-004 (intratumoral; intravenous; intraperitoneal) specifically localizes to orthotopic MMT breast tumors in C57BL/6 mice, with the strongest tumor localization observed via intraperitoneal administration[1].
TAB-004 (1:500-1:1000 dilution; 2-4 μg/mL) specifically detects human MUC1 on pancreatic cancer cells and > 95% of two distinct pancreatic cancer stem cell populations (CD133+ and Triple+) in athymic nude mouse tumor models, and accurately identifies shed MUC1 in serum from mice with MUC1 high tumors[2].
