TP-808 features highly modified stereocenters and functional groups (such as dimethylamino, TBS-protected phenoxy, benzyloxy, and isoxazole rings). It can rapidly construct the tetracyclic ABCD core skeleton of tetracyclines via Michael-Dieckmann condensation with a D-ring precursor, and serves as an essential building block for achieving "convergent total synthesis of tetracyclines and breaking through the bottleneck of semi-synthetic modification"[1].
