Trimethylacetaldehyde (Pivalic aldehyde; Pivalaldehyde) is an inhibitor of E. coli betaine aldehyde dehydrogenase (ALDH) with a Ki of 4.5 mM. Trimethylacetaldehyde serves as a substrate for Drosophila melanogaster alcohol dehydrogenase (AdhS), undergoing oxidation and disproportionation reactions with the release of NADH, and also forms an inactive binary enzyme-aldehyde complex via a compulsory ordered reaction mechanism.

Biological Activity
Trimethylacetaldehyde (0.5-10 mM) is oxidized by purified Drosophila melanogaster AdhS in a dismutation reaction at pH 9.5 and 23.5 °C, with a Km of 2.4 mM[1].
Trimethylacetaldehyde (100 mM) competitively inhibits purified betaine aldehyde dehydrogenase from Escherichia coli MC4100 (pFF423), with an apparent inhibition constant of 4.5 mM, and reduces the enzyme activity to 11% of that in the control group at a concentration of 100 mM[2].
