KIO-301
KIO-301 is a photosensitizer with selective retinal ganglion cell uptake properties. KIO-301 acts on HCN channels and endogenous voltage-gated cation channels, thereby enabling photoswitch-mediated photoresponses and light-driven action potential generation. KIO-301 confers rapid and reliable light-evoked spike responses in retinal explants from mouse models of retinal degeneration, producing sustained light-evoked responses. KIO-301 can be used in research related to retinal degeneration.

Biological Activity
Description
In Vitro
KIO‑301 enters RGCs through the P2X7 large-pore channel highly expressed in degenerated retinas, non-covalently binds to the intracellular domain of HCN channels, and relies on light-controlled reversible isomerization to regulate HCN channel opening and closing and modulate Na+ influx, thereby controlling RGC depolarization and action potential firing[2].
