Leucyl-phenylalanine is a linear dipeptide composed of L-leucine and L-phenylalanine, and also serves as a cyclization substrate. Leucyl-phenylalanine is an incomplete degradation product generated during protein breakdown or catabolism. Leucyl-phenylalanine undergoes heat-induced solid-state cyclization via an autocatalytic mechanism to form its cyclic diketopiperazine analog.

In Vitro
Leucyl-phenylalanine (Leu-Phe) undergoes solid-state cyclization, which is an autocatalytic reaction[1].
Leucyl-phenylalanine exists in neutral form in amorphous films and in zwitterionic form in crystalline films; heating to 100-120 °C triggers a cyclization reaction via conversion to the neutral form, and the cyclization reaction completes upon heating to 140 °C[1].
Leucyl‑phenylalanine smoothes its initial flocculent structure upon heating to 110 °C, and further heating to 150 °C triggers the generation of fibrous structures[1].
