Ac-Trp-Glu-His-Asp-AMC (AC-WEHD-AMC) is a fluorescent peptide caspase-1 substrate. Ac-Trp-Glu-His-Asp-AMC is cleaved by caspase-1, enabling the measurement of caspase-1 proteolytic activity. Ac-Trp-Glu-His-Asp-AMC is cleaved by proinflammatory caspases but not by proapoptotic caspases. Ac-Trp-Glu-His-Asp-AMC requires higher enzyme concentrations to detect active caspase-1.

Description
In Vitro
Ac-WEHD-AMC (10 nM) exhibits selectivity similar to that of CM-269, being cleaved by inflammatory caspase-1, -4, and -5, but not by apoptotic caspases[2].
Ac-DEVD-AFC (10 μM; 45 s UVC, 18 hr incubation) detects significant activation of pro-apoptotic caspases in UV-irradiated HeLa cell lysates[2].
Ac-WEHD-AMC (20 μM; 60 min) serves as a fluorescent substrate for detecting NLRP1-driven caspase-1 activation in recombinant inflammasome assays[3].
