AMG-329
AMG-329 (MEDI1116) is a human immunoglobulin G1λ monoclonal antibody targeting FLT3L. AMG-329 selectively binds to FLT3L, blocks FLT3L-FLT3 interaction, neutralizes soluble and cell-bound human FLT3L activity. AMG-329 reduces circulating relative proportions of plasmacytoid dendritic cells, conventional dendritic cells, and classical monocytes. AMG-329 does not induce antibody dependent cellular cytotoxicity. AMG-329 can be used for the research of sjögren’s disease.

AMG-329 neutralizes soluble human FLT3L in RS4;11 cells, with no activity against mouse or rat FLT3L[1].
AMG-329 neutralizes cell-bound human FLT3L in CD4+ T cell-RS4[1].
AMG-329 (10-3-102 μg/mL; 6 h) does not induce ADCC in IL-7-treated human CD4+ T cells co-cultured with allogeneic PBMCs[1].
AMG-329 (0.001-100 μg/mL; overnight) does not bind purified human C1q[1].
AMG-329 is specific to FLT3L and does not interact with other class III tyrosine kinase receptor ligands including SCF, M-CSF, and PDGF[1].

Result:Induced significant decreases in plasmacytoid dendritic cells (pDCs) relative to baseline vs control starting on day 8 of the 5-week study, with maximal decreases between days 25 and 81 of recovery.
Induced significant decreases in conventional dendritic cell 1 (cDC1) relative to baseline vs control in the 5-week study, with depletion maintained through day 109 of recovery.
Induced significant decreases in conventional dendritic cell 2 (cDC2) relative to baseline vs control in the 5-week study, maintained through day 81 of recovery.
Induced significant decreases in classical monocytes relative to baseline vs control on day 29 of the 5-week study.
Caused all affected cell subsets to generally return to baseline levels by the end of the recovery phase for both studies.
Showed no significant changes in nonclassical monocytes, intermediate monocytes, or B cells.
Resulted in no differences in anti-keyhole limpet hemocyanin (KLH) IgM or IgG titers between treated and control animals.
