BODIPY TR methyl ester
BODIPY TR methyl ester is a lipophilic GFP Counterstain. BODIPY TR methyl ester dye readily permeates cell membranes and localizes in endomembranous organelles but not localize strongly in plasma membranes. BODIPY TR methyl ester is an excellent red fluorescent vital dye (Ex=568 nm, Em=625 nm), can be used to reveal the location and shapes of cell nuclei, the shapes of cells within embryonic tissues, as well as the bound aries of organ-forming tissues within the whole embryo.

Biological Activity
Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
Vital staining procedures[1]:
1. Dissolve BODIPY TR methyl ester in anhydrous dimethyl sulfoxide (DMSO) to prepare a 5 mM stock solution.
2. Dilute the DMSO stock solution in embryo rearing medium (ERM) buffered with 5 mM HEPES (pH 7.2) to prepare a BODIPY TR methyl ester labeling solution with a final concentration of 100 μM and a DMSO volume fraction of 2%.
3. Stain the embryos in 100 μM BODIPY TR methyl ester solution for 1 h, then wash them three times successively in HEPES-buffered ERM.
4. Mount the embryos in an open-faced chamber for imaging.
5. For embryos used in fixation experiments, fix them in HEPES-buffered ERM containing 4% paraformaldehyde at 4°C for 1 h, then wash with ERM and mount for confocal imaging.
6. Analyze the sample under a confocal microscope (Ex = 568 nm, Em = 625 nm).
