Description

Bromocresol purple sodium is a pH indicator, whose color changes from yellow to purple when the pH increases from 5.2 to 6.8. Bromocresol purple sodium can be used for detecting yeast cells with plasma membrane damage and measuring serum albumin concentrations[1][2][3].

In Vitro

Arabidopsis thaliana (L.) seedlings are transferred to Murashige and Skoog (MS) or low phosphorus medium which contained 0.75% [weight/volume (w/v)] agar, 0.006% (w/v) Bromocresol purple sodium, pH 5.8 for 8-24 h. There are no significant color changes around the roots of wild-type (WT) seedlings on the MS medium[1].
Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
1. The yeast cells were centrifuged, washed with distilled water and resuspended in 5mM-citrate-phosphate buffer, pH 4.6.
2. Aliquots containing 107 cells were suspended in the same buffer, supplemented with glucose to a final concentration of 220 mM and different volumes of killer toxin preparation were added so that the final volume was 1 mL.
3. Incubation with the killer was then carried out in Eppendorf vials at 25°C on a shaker for 120 min.
4. 50 μL 10 mM Bromocresol purple sodium in 5 mM-citrate-phosphate buffer, pH 4.6, was added to the samples and incubation continued for 60 min under the same conditions.
5. The samples were then centrifuged, thoroughly washed with 5 mM-citrate-phosphate buffer.
6. The fraction of stained cells was determined in a-Jenalumar fluorescence microscope using violet light for excitation. Cell fluorescence was observed using filter G 251, which cuts off wavelengths below 450 nm.
