DOPE-PEG2000-RVG29 is a functionalized PEGylated lipid conjugate composed of three modular components: dioleoylphosphatidylethanolamine (DOPE) as the lipid anchoring group, a polyethylene glycol (PEG) spacer arm with a molecular weight of 2000 Da, and RVG29 peptide (YTIWMPENPRPGTPCDIFTNSRGKRASNG) (HY-P5623) as the terminal targeting/functional ligand. DOPE-PEG2000-RVG29 can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against the nAChR α7 subunit on neurons and brain endothelial cells.

DOPE-PEG2000-RVG29 is specifically designed for active targeting nanomedicine applications. It can be incorporated into liposomes, lipid nanoparticles (LNPs), or other nanocarriers to confer ligand-directed targeting capability against the nAChR α7 subunit on neurons and brain endothelial cells.
Composition and role of each component:
1. PEG2000 (polyethylene glycol, MW 2000): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
2. RVG29 (RVG29 peptide (YTIWMPENPRPGTPCDIFTNSRGKRASNG)): Targeting ligand-a 29-amino acid peptide derived from rabies virus glycoprotein that specifically binds to the nicotinic acetylcholine receptor (nAChR) α7 subunit, which is abundantly expressed on neuronal cells. Enables transcytosis across the blood-brain barrier (BBB) and targeted delivery to the central nervous system.
