TREM2 agonist-3 (Compound 4i) is a TREM2 agonist with a KD value of 19.0 µM. The KD value of TREM2 agonist-3 for TREM1 is 39.8 µM. TREM2 agonist-3 induces an increase in phosphorylated SYK levels. TREM2 agonist-3 can be used in the research of neurodegenerative diseases and other diseases associated with TREM2 dysfunction.

TREM2 agonist-3 (10 min) binds to recombinant TREM2 with a Kd value of 19 μM[1].
TREM2 agonist-3 (10-100 μM; 1 h) does not reduce the viability of human HMC3 microglial cells following 1-hour treatment at concentrations of 10, 30 or 100 μM, as determined by MTS assay[1].
TREM2 agonist-3 (25 μM; 1 h) activates the TREM2/DAP12 signaling pathway in HEK293-hTREM2/DAP12 cells. AlphaLISA assays show that the level of phosphorylated Syk (Tyr525/526) increases after treatment with 25 μM TREM2 agonist-3 for 1 hour[1].
TREM2 agonist-3 (25 μM; 30 min or 1 h) enhances the phagocytosis of opsonized fluorescent latex microspheres by mouse BV2 microglial cells, increasing the proportion of phagocytic cells by approximately 46%[1].

Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline

This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 5 mg/mL (12.55 mM); Clear solution
This protocol yields a clear solution of ≥ 5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (50.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
