DMG-PEG3400-WYRGRL is a functionalized PEGylated lipid conjugate composed of three modular components: dimyristoylglycerol (DMG) as the lipid anchoring group, a polyethylene glycol (PEG) spacer arm with a molecular weight of 3400 Da, and WYRGRL peptide (HY-P10739)as the terminal targeting/functional ligand. DMG-PEG3400-WYRGRL is specifically designed for active targeting nanomedicine applications. Composition and role of each component:
1. DMG (dimyristoylglycerol): Lipid anchoring group-a diacylglycerol with two C14 chains, providing moderate membrane anchorage; its short-chain, high-fluidity characteristics enable faster dissociation of PEG-lipids from nanoparticle surfaces, facilitating endosomal escape and cellular uptake.
2. PEG3400 (polyethylene glycol, MW 3400): Hydrophilic polymer spacer arm that forms a steric corona around nanoparticles, reducing opsonization and reticuloendothelial system (RES) clearance, prolonging circulation half-life, and providing spatial separation between the lipid surface and targeting ligand.
3. WYRGRL (WYRGRL peptide): Targeting ligand-a targeting peptide with the sequence tryptophan-tyrosine-arginine-glycine-arginine-leucine, designed to enhance tissue-specific binding. The WYRGRL motif has been explored for targeting specific cell populations in tumor and inflammatory microenvironments, utilizing its arginine-rich sequence to simultaneously achieve cell penetration and targeting functions.
