DOPE-PEG2000-Asp6

DOPE-PEG2000-Asp6: surface functionalization of liposomes and nanoparticles for bone-targeted drug delivery, applicable in treatment of bone metastasis, osteoporosis, bone fractures, bone infections (osteomyelitis), and other skeletal diseases.
Structural composition:
(1) DOPE; a fusogenic phospholipid with two oleoyl chains that anchors into lipid bilayers and promotes membrane fusion for cellular uptake;
(2) PEG2000; a hydrophilic polymer that provides steric stabilization, reduces opsonization and immune clearance, and extends circulation half-life;
(3) Asp6 peptide; hexa-aspartic acid, a repeating sequence of six aspartic acid residues that mimics the acidic domains of bone-binding proteins (e.g., osteocalcin, bone sialoprotein) and specifically binds to hydroxyapatite (the calcium phosphate mineral matrix of bone), directing nanocarriers to bone tissue surfaces for localized drug delivery to skeletal sites with high affinity and selectivity.
