Description

FRETS-VWF73, a 73-amino-acid peptide, is a fluorogenic substrate for ADAMTS13 assay (Ex = 340 nm; Em = 450 nm). FRETS-VWF73 is a predictive tool for thrombotic thrombocytopenic purpura[1][2].

In Vitro

Guidelines (The following are our recommended solutions. These are only guidelines and should be modified according to your specific needs)
1.1 Preparation of storage solution
FRETS-VWF73 is dissolved in DMSO to prepare 1 mM FRETS-VWF73 stock solution.
Note: FRETS-VWF73 stock solution is recommended to be aliquoted and stored at -20°C or -80°C in the dark.
1.2 Preparation of working solution
Dilute the FRETS-VWF73 stock solution to 4 μM with assay buffer.
Note: Please adjust the concentration of FRETS-VWF73 working solution according to the actual situation, and prepare it fresh each time you use it.
1.3 Experimental procedures
1) Pooled human plasma (a range of 0-8 μL as a standard), or 4 μL of each test plasma, is diluted in 100 μL of assay buffer (5 mmol/L Bis-Tris, 25 mmol/L CaCl2, 0.005% Tween-20, pH 6) in a 96-well white plate.
2) Then, 100 μL of 4 μM FRETS-VWF73 in the assay buffer is added to each well.
3) Fluorescence is measured at 30℃ in a Wallac 1420 ARVO multilabel counter equipped with a 340-nm excitation filter and a 450-nm emission filter.
4) Fluorescence is measured every 5 min. The reaction rate is calculated by linear regression analysis of fluorescence over time from 0 to 60 min using the prism software.
