DOPE-PEG2000-TH: surface functionalization of liposomes and nanoparticles for thrombus-targeted drug delivery and thrombus imaging, applicable in thrombolytic therapy, anticoagulant delivery, and diagnosis of thrombotic diseases.
Structural composition:
(1) DOPE; a phospholipid with two unsaturated oleoyl chains that anchors into lipid bilayers and provides fusogenic activity for cellular uptake;
(2) PEG2000; a hydrophilic polymer that creates a steric shield reducing opsonization and RES clearance, prolonging circulation time;
(3) TH peptide; a thrombus-homing peptide that specifically binds to components of blood clots (such as fibrin networks or activated platelet surface receptors like GPIIb/IIIa), directing nanocarriers to thrombotic sites for targeted delivery of thrombolytic agents (e.g., tPA, urokinase) or anticoagulants, enabling site-specific clot dissolution with reduced bleeding risk.
