6:2 Cl-PFAES (0.04-1.0 mg/kg/d; p.o.; daily; 56 days) reduces relative testis weight by 8% and absolute/relative epididymis weights by 14% and 16%, respectively, at the highest dose of 1.0 mg/kg/d, but does not alter reproductive hormones, histopathology, sperm quality, fertility, or testicular gene/protein expression in adult male BALB/c mice[2].

Altered transcriptional levels of multiple hypothalamic-pituitary-gonadal (HPG) axis genes in F0 zebrafish at 5 and 50 μg/L, with down-regulation of brain genes (e.g., gnrh3, fshb, lhb) in most cases, and altered gonadal steroidogenic gene expression (e.g., increased cyp19a in testes, increased hmgra, star, cyp17 in ovaries).
Induced 1576 differentially expressed genes in testes of F0 zebrafish at 50 μg/L, enriched in pathways related to spermatogenesis (e.g., cytokine-cytokine receptor interaction, lysosome, focal adhesion).
Significantly reduced body weight, body length, and female GSI in F1 offspring from 5 μg/L exposed F0 parents; reduced serum testosterone levels in male F1 offspring.
Increased embryo malformation rate to 9.0% and 15.0% at 120 hpf, and increased mortality rates at 120 hpf in F1 offspring from 5 and 50 μg/L exposed F0 parents; F1 offspring from 50 μg/L exposed F0 parents died before 15 dpf.
Detected target reagent in adult blood (0.31 ng/mL males, 0.37 ng/mL females) and gonads (0.10 ng/mg males, 0.12 ng/mg females), and reduced mature oocytes in ovaries of F1 offspring from 5 μg/L exposed F0 parents.
Increased embryo malformation rate to 5.0% at 120 hpf and reduced survival rate in F2 offspring from 5 μg/L exposed F0 parents.

Reached epididymis 6:2 Cl-PFAES levels of 1.69 μg/g (0.04 mg/kg/d), 2.68 μg/g (0.2 mg/kg/d), and 9.64 μg/g (1.0 mg/kg/d), representing 26.1-, 41.4-, and 149-fold increases over control (0.065 μg/g).
Showed no significant changes across all doses in serum or testicular testosterone, estradiol, follicle-stimulating hormone, or luteinizing hormone levels; testis/epididymis histopathology; sperm count; fertility metrics (mating index, litter size, pup birth/weaning weight, pup sex ratio); testicular gene expression profiles; or testicular levels of steroidogenic proteins (STAR, CYP11A1, CYP17A1, 3β-HSD) or cell junction proteins (occludin, β-catenin, connexin 43).

- For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
- To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for In Vivo experiments, it is recommended to prepare freshly and use it on the same day.
- The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.
Add each solvent one by one: 10% DMSO 40% PEG300 5% Tween-80 45% Saline
Solubility: ≥ 2.5 mg/mL (4.38 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 400 μL PEG300, and mix evenly; then add 50 μL Tween-80 and mix evenly; then add 450 μL Saline to adjust the volume to 1 mL.
Preparation of Saline: Dissolve 0.9 g sodium chloride in ddH₂O and dilute to 100 mL to obtain a clear Saline solution.
Add each solvent one by one: 10% DMSO 90% (20% SBE-β-CD in Saline)
Solubility: ≥ 2.5 mg/mL (4.38 mM); Clear solution
This protocol yields a clear solution of ≥ 2.5 mg/mL (saturation unknown).
Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (25.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.
