hIAPP22–27
hIAPP22-27 is a hexapeptide corresponding to residues 22-27 of human islet amyloid polypeptide (hIAPP) and is the key amyloid core sequence of hIAPP. hIAPP22-27 can self-assemble under neutral pH conditions to form β-sheet-containing amyloid fibrils and is cytotoxic to pancreatic β cells. hIAPP22-27 can be used in research related to type 2 diabetes.

hIAPP22-27 can self-assemble into amyloid fibrils upon incubation with orbital shaking in neutral pH buffer, and small-angle X-ray scattering revealed that the fibrils exhibit a ribbon-like structure[1].

The following protocol uses the MTT colorimetric assay as a classic literature-established method for assessing cell viability/metabolic activity in cultured mammalian cells. MTT[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] is reduced by metabolically active cells to a colored formazan product; the amount of formazan is quantified spectrophotometrically and provides an indirect measure of metabolically active viable cells. Importantly, MTT reduction reflects cellular oxidoreductase/metabolic activity rather than an absolute direct count of living cells, so changes in cellular metabolism can alter the signal independently of cell number.
