DOPE-PEG2000-GRGDS
DOPE-PEG2000-GRGDS (DOPE-PEG2000-Mal-Cys-GRGDS) is an amphiphilic lipid-PEG-peptide conjugate consisting of DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine), PEG2000, and the GRGDS peptide (Gly-Arg-Gly-Asp-Ser), an RGD motif-containing peptide. GRGDS peptide specifically binds to integrin receptors (particularly alphavbeta3 and alphavbeta5) that are overexpressed on tumor cells and angiogenic endothelial cells. DOPE-PEG2000-GRGDS is used for integrin-targeted drug delivery and anti-angiogenic research.

In Vitro
DOPE-PEG2000-GRGDS: surface functionalization of liposomes and nanoparticles for integrin receptor-targeted drug delivery to tumors and angiogenic vasculature, applicable in cancer therapy and anti-angiogenic treatment.
Structural composition:
(1) DOPE; a fusogenic phospholipid with two oleoyl chains that anchors into lipid bilayers and promotes pH-dependent membrane fusion for intracellular drug release;
(2) PEG2000; a hydrophilic polymer that provides steric stabilization, reduces protein adsorption and immune clearance, and prolongs circulation half-life;
(3) GRGDS peptide; an RGD-containing peptide that specifically binds to integrin receptors (alphavbeta3, alphavbeta5) overexpressed on tumor cells and activated endothelial cells, enabling active targeting of tumor tissues and angiogenic vasculature while potentially exerting anti-angiogenic effects by blocking integrin-mediated signaling.

Endothelial Tube Formation Assay
Endothelial tube formation assay evaluates the ability of endothelial cells to attach, migrate, align, and organize into capillary-like networks when cultured on gelled basement membrane extract or Matrigel; the readout is the morphology and quantity of tube-like networks, which reflects an in vitro endothelial morphogenesis step related to angiogenesis. Basement membrane extract/Matrigel provides laminin-rich extracellular matrix cues that support endothelial differentiation into capillary-like structures, but it can contain biologically active growth factors, so growth-factor-reduced matrix is preferred when testing defined angiogenic stimulators or inhibitors.
