X-34 is a lipophilic and bright yellow-green fluorescent derivative of Congo red (HY-D0236). X-34 can be used to stain neuritic and diffuse plaques, neurofibrillary tangles (NFTs), neuropil threads, and cerebrovascular amyloid in the brain. X-34 can be used for research of Alzheimer’s disease.

Biological Activity
Guide (The following is our recommended protocol. This protocol is only a guide and should be modified according to your specific needs).
1. Sample preparation[1]
1.1 Paraformaldehyde-fixed tissue was cryoprotected by sequential incubation in 15% and 30% sucrose (in 0.1 M phosphate buffer), frozen, and cryosectioned.
1.2 Frozen sections were collected in Falcon 6-well plates containing
0.1 M phosphate buffer, in a 1:6 series.
1.3 Sections are stored at 4°C until assayed.
1.4 Before staining, the sections are washed for 5 min in PBS.
2. X-34 General Staining Procedure[1]
2.1 Quenched or unquenched tissue sections are taken from PBS.
2.2 Add into a solution of X-34 in 40% ethanol/60% distilled H2O
(adjusted to pH 10 by addition of 1 N NaOH) at the appropriate concentration (100 nM to 1 mM) for the appropriate
period of time (0.5-10 min).
2.3 The sections are then dipped briefly five times in tapwater before differentiation in 0.2% NaOH in 80% ethanol for 2 min.
2.4 The sections are then placed in tapwater for 10 min for microscopic observation.
