6-NBDG is a fluorescent glucose analogue, it can be used for fluorescence imaging and monitoring glucose transport and uptake. 6-NBDG can be used as a fluorescent probe for detecting macrophage-rich atherosclerotic plaques.

Biological Activity
Guide (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).
1. Cell Preparation
Culture cells to appropriate confluence on suitable imaging dishes or plates.
2. Glucose Starvation
Wash cells 2-3 times with warm PBS (without Mg2?/Ca2?) to remove existing glucose.
Incubate cells in glucose-free DMEM or KRH buffer for 30-60 minutes at 37°C to increase transporter activity.
3. 6-NBDG Incubation
Replace medium with glucose-free medium containing 50-150 μM 6-NBDG.
Incubate at 37°C for 15-60 minutes, protecting the samples from light (e.g., aluminum foil).
4. Washing and Measurement
Remove the 6-NBDG solution.
Wash cells 3 times with ice-cold PBS to remove non-specific fluorescence.
Measure immediately using a fluorescent plate reader, flow cytometer, or confocal microscope (Ex: 465-488 nm / Em: 530-542 nm).
