LL-340001 is the receptor-active metabolite released after hydrolysis of Elunetirom (HY-160207) by FAAH in the central nervous system. LL-340001 acts as a thyroid hormone receptor (TR) agonist, with potent full agonist activity at both TRα and TRβ. LL-340001 exhibits neuronal plasticity activity and can activate programs related to neuroplasticity and mitochondrial biogenesis in neuronal systems. LL-340001 can be used in the research of major depressive disorder and bipolar depression.

In Vitro
LL-340001 (administered for 23 h) potently activates TRα and TRβ receptors in HEK293-derived reporter cells[1].
LL-340001 (1-30 nM, 3 μM; 72 h) increases the number of MAP-2-positive neurons, neurite length, number of branches, and number of neurite terminals in primary cortical neurons from immature rats within 72 hours, and exhibits potent activity in the low nanomolar range[1].
The increase in the number of MAP-2-positive neurons induced by LL-340001 (3-30 nM; 73 h) in immature primary rat cortical neurons is sensitive to TrkB inhibition but independent of the 5-HT2A signaling pathway, whereas its neuritogenic effect is not consistently sensitive to TrkB antagonism[1].
LL-340001 (1 nM-1 μM, 28 h) maintains the number of synapses in mature primary rat hippocampal neurons under pathological stress induced by Aβ1-42, and exhibits potent activity within the low nanomolar range[1].
LL-340001 (0.3-3 nM; 72 h) enhances signaling pathways associated with mitochondrial biogenesis (upregulates nuclear PGC-1α and NRF2 expression), increases the number of functional mitochondria, and elevates ATP levels in mature primary rat hippocampal neurons within 72 h, exhibiting potent activity in the low nanomolar range[1].
