MAUra-DTB
MAUra-DTB is a 1-methyl-4-arylurazole (MAUra) chemical probe conjugated with desthiobiotin, which preferentially labels tyrosine residues exposed on protein surfaces under photocatalytic or laccase-catalyzed conditions (). The desthiobiotin moiety of MAUra-DTB supports affinity enrichment based on streptavidin or NeutrAvidin. MAUra-DTB enables proteome-wide labeling in cell lysates, and after enrichment of labeled peptides by NeutrAvidin, the labeling sites can be identified via nanoLC-MS/MS. MAUra-DTB can be used for photocatalytic proximity labeling, target protein identification and proteomic studies.

Biological Activity
MAUra-DTB (500 μM; 30 min) selectively labels nuclear proteins in proximity to HaloTag-H2B transiently expressed in HeLa cells, with localization confirmed via colocalization of streptavidin-Texas Red and nuclear Hoechst 33342 signals[2].
MAUra-DTB (500 μM; 10-60 min) preferentially labels highly surface-exposed tyrosine residues in A431 cell lysate proteomes, with 2017 single-modification sites identified after 10 minutes and over 3000 single-modification sites identified after 60 minutes, showing minimal tryptophan cross-reactivity[3].
