AGS-1C4D4 displays potent ADCC activity against PSCA-expressing HPAC and Panc0203 pancreatic cell lines, with activity greatly reduced by deglycosylation, and is more potent than the hybridoma-derived AGS-PSCA antibody in this assay[1].
AGS-1C4D4 exhibits CDC activity against PSCA-expressing recombinant B300.19 cells, with activity greatly reduced by deglycosylation, and is equally effective as the hybridoma-derived AGS-PSCA antibody in this assay[1].
Intact AGS-1C4D4 exhibits no direct cytotoxic activity against HPAC pancreatic cells in vitro[1].
AGS-1C4D4 binds to PSCA with a Kd of 2.0 × 10-10 M, displaying similar binding affinity to the hybridoma-derived AGS-PSCA antibody[1].

In Vivo

AGS-1C4D4 significantly inhibits established pancreatic tumor growth and metastasis in orthotopic HPAC xenograft models when combined with Gemcitabine, and also significantly inhibits pancreatic tumor formation, local invasion, and distant metastasis in the same orthotopic xenograft model[1].

Biological Activity
  • Flow Cytometry analysis of HepG2 cells with MK-4721 (HY-P991229, red) . Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then cells were stained with the primary antibody at 1/200 dilution for an hour at 4℃. Goat Anti-Human IgG H&L (AF488) (HY-P83776) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Human IgG1 kappa(HY-P99001, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
