NSC156565 is a Pycsar immune system inhibitor and phage infection potentiator. NSC156565 blocks PycTM cytotoxic effects via prevention of 3′,5′-cCMP-mediated activation or inhibition of activated effector membrane-perturbing activity. NSC156565 enables bacteriophage Bas34 to overcome Pycsar-mediated resistance, facilitating productive phage infection and replication.

Biological Activity
NSC156565 (11-111 μM; 1 h pretreatment) potently inhibits the Pycsar antiphage defense in Pycsar-expressing E. coli JW5503ΔtolC, enabling >104-fold increased Bas34 phage replication and phage-induced bacterial lysis when used at 111 μM[1].
NSC156565 (111 μM) does not inhibit cCMP production by the PycC cyclase in PycC-expressing E. coli MG1655[1].
NSC156565 (11 μM; 200 min) blocks PycTM-induced cell death in Pycsar-expressing E. coli JW5503ΔtolC, rescuing growth inhibited by exogenous cCMP[1].
NSC156565 (0.01-100 μM; 1 h pretreatment) inhibits the Pycsar antiphage defense in Pycsar-expressing E. coli JW5503ΔtolC in a dose-dependent manner[1].
