Oxyphenonium bromide is an orally effective competitive muscarinic acetylcholine receptor (mAChR) antagonist. Oxyphenonium bromide competitively binds to and blocks mAChR, thereby inhibiting the parasympathetic pathway. By inhibiting CHRM4, Oxyphenonium bromide increases cAMP levels and activates the CREB transcriptional program, thereby promoting erythroid progenitor cell expansion. Oxyphenonium bromide can be used in research on asthma, anemia, and gastric ulcers.

In Vitro
Oxyphenonium bromide (200 μM; 14 days) can expand erythroid cells and increase BFU-E colony formation in human CD34+ HSPCs[2].
Oxyphenonium bromide (100 μM; 3-9 days) promotes long-term cell expansion, self-renewal, and colony formation in mouse early erythroid progenitor cells (BFU-E)[2].
Oxyphenonium bromide (100 μM; 45 min) increases phosphorylated CREB in mouse fetal liver BFU-Es[2].

In Vivo
Oxyphenonium bromide (25 mg/kg; i.p.; once daily; 45 days) corrects anemia and promotes BFU-E self-renewal in the Mx1-Cre Srsf2P95H/WT myelodysplastic syndrome (MDS) C57bl/6 mouse model[2].
Oxyphenonium bromide (21.4 mg/kg; s.c.; single administration; 15 min) inhibited the absorption rate of the radiopharmaceutical (DF32P) in a subcutaneous absorption model in albino mice[4].
Oxyphenonium bromide (0.4 mg/kg; i.v.; single administration; short-term observation) does not alter EEG activity and does not block amphetamine-induced EEG activation responses in the EEG activation model[5].
Oxyphenonium bromide (5 mg/kg; i.m.; twice daily; 7 and 14 days) reduces ulcer area, promotes epithelialization and collagen formation, and modulates enzyme activity in the ulcer region in a rat model of gastric ulcer[7].
