TRAIL R1/TNFRSF10A Protein, Human (HEK293, His-Avi)

Cat. No.: HY-P78529
Handling Instructions

TRAIL R1/TNFRSF10A Protein, a receptor for TNFSF10/TRAIL, initiates apoptosis by recruiting caspase-8 via FADD, forming the death-inducing signaling complex (DISC). It activates NF-kappa-B and interacts with TRADD and RIPK1 in its monomeric state. TRAIL R1/TNFRSF10A forms homooligomers and heterooligomers with TNFRSF10B, interacting with the TNFSF10 homotrimer. It also interacts with ARAP1 and ZDHHC3, highlighting its role in complex signaling networks. In the absence of stimulation, it interacts with BIRC2, DDX3X, and GSK3B, and stimulation enhances these interactions, accompanied by cleavage of DDX3X and BIRC2, emphasizing its multifaceted role in apoptotic and signaling pathways. TRAIL R1/TNFRSF10A Protein, Human (HEK293, His-Avi) is the recombinant human-derived TRAIL R1/TNFRSF10A protein, expressed by HEK293, with C-Avi, C-His labeled tag. The total length of TRAIL R1/TNFRSF10A Protein, Human (HEK293, His-Avi) is 206 a.a., with molecular weight of 25-30 kDa.

Background

The TRAIL R1/TNFRSF10A Protein serves as a receptor for the cytotoxic ligand TNFSF10/TRAIL. Upon activation, the adapter molecule FADD recruits caspase-8 to the receptor, forming the death-inducing signaling complex (DISC), leading to caspase-8 proteolytic activation and initiating the subsequent cascade of caspases, mediating apoptosis. Additionally, TRAIL R1/TNFRSF10A promotes the activation of NF-kappa-B. In its monomeric state, it can interact with TRADD and RIPK1. Moreover, TRAIL R1/TNFRSF10A forms homooligomers and heterooligomers with TNFRSF10B, and three TRAIL R1 molecules interact with the TNFSF10 homotrimer. The receptor also interacts with ARAP1 and ZDHHC3, further highlighting its involvement in complex signaling networks. In the absence of stimulation, TRAIL R1/TNFRSF10A interacts with BIRC2, DDX3X, and GSK3B, and this interaction is enhanced upon receptor stimulation, accompanied by cleavage of DDX3X and BIRC2. These intricate interactions emphasize the multifaceted role of TRAIL R1/TNFRSF10A in apoptotic and signaling pathways.

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.

The reconstitution calculator equation

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

Volume (to add to vial) = Mass (in vial) ÷ Desired Reconstitution Concentration

The dilution calculator equation

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)

This equation is commonly abbreviated as: C1V1 = C2V2

Concentration (start) × Volume (start) = Concentration (final) × Volume (final)
× = ×
C1   V1   C2   V2

The specific activity calculator equation

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)

Specific Activity (Unit/mg) = 106 ÷ Biological Activity (ED50)
Unit/mg = 106 ÷ ng/mL
