The TRIB3 protein, an inactive kinase, operates as a pivotal regulator of the integrated stress response (ISR), a mechanism crucial for adapting to diverse stress conditions. It functions as an inhibitor of the transcriptional activity of DDIT3/CHOP and plays a role in DDIT3/CHOP-dependent cell death during endoplasmic reticulum (ER) stress, particularly in the context of programmed neuronal cell death. Acting as a negative feedback regulator within the ISR, TRIB3 interacts with ATF4, inhibiting ATF4 transcriptional activity while being promoted by ATF4 expression. Moreover, TRIB3 disrupts insulin signaling by directly binding to Akt kinases, impeding their activation, and may mask the 'Thr-308' phosphorylation site in AKT1. Additionally, TRIB3 interacts with various signaling molecules, including NF-kappa-B transactivator p65 RELA, MAPK kinases, and APOBEC3 proteins, showcasing its multifaceted role in diverse cellular processes and stress responses.
